There is no 'Suspend Run' feature — you can simply quit whenever you want and the game will remember where you are when you go back in.

What Happens if You Quit Saros Mid-Run?

If you close Saros while you're half way through a run, you will be able to resume from wherever you left off.

You don't need to manually save or select any options in the menus.

If you want to double check, pause the game and go to Game Slots. You will be presented with three save slots, and your current one will be highlighted. It will tell you the last time the game saved to that slot, and it's always very up-to-date as it autosaves very regularly.
